实验性肝纤维化时MMP-1、TIMP-1的表达与Ⅰ、Ⅲ型胶原含量变化的关系

To observe the relativity of expression of MMP-1、TIMP-1 and variability of type Ⅰ and Ⅲ collagen during experimental liver fibrosis, we made the rats liver fibrosis model by immune attack of human serum albumin (HSB) , and detected the expression of MMP-1 and TIMP-1 by quantitative RT-PCR, assayed the type I and type Ⅲ collagen by immunohistochemistry, then analyzed relativity of expression of MMP-1、TIMP-1 and variability of typeⅠ、Ⅲ collagen during liver fibrosis We found that the change of content of type Ⅰ and type Ⅲ collagen presented a high relativity (r=0 875, P<0 001) ;no difference was found between normal group and fibrosis group in the expression of MMP-1, and no relativity was found between the expression of MMP-1 and the change of content of type Ⅰ、type Ⅲ collagen;but we found the expression of TIMP-1 became significantly high after formation of liver fibrosis (P<0 01) , and it has the high relativity to the variability of type Ⅰand Ⅲ collagen So we got the conclusion that the expression of MMP-1 has no significant change during liver fibrosis, but that of TIMP-1 become obvious high, and there is a distinct relativity between the expression of TIMP-1 and the increasing of type Ⅰ、type Ⅲ collagen during liver fibrosis All of these suggest that increasing of the expression of TIMP-1 play a important role in the formation of liver fibrosis
